  [摘要] 該文建立一種同時(shí)測(cè)定4種蟲(chóng)草類(lèi)藥材(冬蟲(chóng)夏草、蛹蟲(chóng)草、蟲(chóng)草菌絲體和蟬花)中蟲(chóng)草素(3′-脫氧腺苷)和2′-脫氧腺苷含量的在線固相萃取-高效液相色譜分析方法。供試品經(jīng)ZORBAX SB-AQ(4.6 mm×12.5 mm,5 μm)色譜柱富集,水-甲醇(91∶9)洗脫;ZORBAX SB-AQ(4.6 mm×150 mm,5 μm)色譜柱分離,流動(dòng)相為0.1%甲酸水溶液-甲醇(91∶9),檢測(cè)波長(zhǎng)260 nm,柱溫40 ℃,流速1.0 mL·min-1,分別對(duì)4種蟲(chóng)草類(lèi)藥材中蟲(chóng)草素和2′-脫氧腺苷含量進(jìn)行檢測(cè)。結(jié)果表明冬蟲(chóng)夏草、蟲(chóng)草菌絲體和蟬花樣品中僅檢測(cè)到2′-脫氧腺苷,蛹蟲(chóng)草樣品中僅檢測(cè)到蟲(chóng)草素,早期報(bào)道的在冬蟲(chóng)夏草中檢測(cè)到的蟲(chóng)草素色譜峰可能是其同分異構(gòu)體2′-脫氧腺苷色譜峰或以2′-脫氧腺苷為主的與蟲(chóng)草素的共洗脫色譜峰。該方法重復(fù)性好,準(zhǔn)確度高,分析時(shí)間短,可用于蟲(chóng)草類(lèi)藥材中蟲(chóng)草素和2′-脫氧腺苷的分析。

  [Abstract] An online SPE-HPLC method for simultaneous determination of cordycepin (3′-deoxyadenosine) and 2′-deoxyadenosine in Cordyceps genus (C. sinensis,C. militaris,Hirsutella sinensis and C. sobolifera) was developed. The samples were eiched on a ZORBAX SB-AQ (4.6 mm×12.5 mm,5 μm) column with isocratic elution by 9% methanol solution. The separation of analytes was performed on a ZORBAX SB-AQ (4.6 mm×150 mm,5 μm) column with gradient elution by 0.1% formic acid solution and methanol (91∶9). The flow rate was 1.0 mL·min-1. Column temperature was 40 ℃ and detection wavelength was 260 nm. This method has been applied for analysis of different Cordyceps genus. The 2′-deoxyadenosine was detected in C. sinensis,Hirsutella sinensis and C. sobolifera. The cordycepin was detected in C. militaris. In summary,the cordycepin chromatographic peak from C. sinensis in some past reports may be the 2′-deoxyadenosine chromatographic peak or the mixture peak of 2′-deoxyadenosine and cordycepin in which 2′-deoxyadenosine content was higher than cordycepin. The developed method is suitable for analysis of cordycepin and 2′-deoxyadenosine in Cordyceps genus.

  冬蟲(chóng)夏草是麥角菌科真菌冬蟲(chóng)夏草菌Cordyceps sinensis(BerK.)Sacc.寄生在蝙蝠蛾科昆蟲(chóng)幼蟲(chóng)上的子座和幼蟲(chóng)尸體的復(fù)合體,具有補(bǔ)肺益腎,止血化痰的功效[1]。現(xiàn)代藥理學(xué)研究表明,冬蟲(chóng)夏草具有抗腫瘤、免疫調(diào)節(jié)、保護(hù)肝臟、保護(hù)腎臟、保護(hù)心血管、延緩衰老等功效[2-5]。蛹蟲(chóng)草為蛹草真菌C. militaris以蟲(chóng)蛹為寄主或在人工培養(yǎng)基上形成的子實(shí)體,蟲(chóng)草菌絲體為中華被毛孢真菌Hirsutella sinensis通過(guò)液體發(fā)酵技術(shù)生產(chǎn)的菌絲體,蟬花為蟬花真菌C. sobolifera寄生于蟬類(lèi)若蟲(chóng)形成的菌蟲(chóng)復(fù)合體[6]。
  蟲(chóng)草素(3′-脫氧腺苷)具有抗腫瘤、抗菌、抗病毒、免疫調(diào)節(jié)、清除自由基等多種藥理作用[7-8],有觀點(diǎn)認(rèn)為其是冬蟲(chóng)夏草的主要活性成分,也有研究認(rèn)為冬蟲(chóng)夏草中蟲(chóng)草素不存在或含量極微,而含有的是2′-脫氧腺苷[9-10]。蟲(chóng)草素與其同分異構(gòu)體2′-脫氧腺苷結(jié)構(gòu)相近,液相分離困難,容易造成錯(cuò)誤指認(rèn)。前期實(shí)驗(yàn)結(jié)果表明蟲(chóng)草素在蛹蟲(chóng)草中含量相對(duì)較高,但在冬蟲(chóng)夏草中含量很低,定量困難,精確指認(rèn)不易[11]。因此有必要開(kāi)發(fā)一種能夠有效分離蟲(chóng)草素和2′-脫氧腺苷,并準(zhǔn)確定量的分析方法,對(duì)蟲(chóng)草類(lèi)藥材中存在的蟲(chóng)草素或2′-脫氧腺苷進(jìn)行定性和定量分析。蟲(chóng)草素和2′-脫氧腺苷在蟲(chóng)草類(lèi)藥材的含量通常較低,本研究通過(guò)固相萃取法對(duì)蟲(chóng)草類(lèi)藥材中的蟲(chóng)草素和2′-脫氧腺苷進(jìn)行富集,進(jìn)而實(shí)現(xiàn)定量分析。
  固相萃。╯olid phase extraction,SPE)是色譜分析中極其重要的一種樣品前處理方法,主要目的是把痕量被測(cè)組分進(jìn)行濃縮、富集、凈化和介質(zhì)轉(zhuǎn)移(把被測(cè)組分從樣品基體中轉(zhuǎn)移到溶劑或氣體中)[12],具有節(jié)省時(shí)間,溶劑用量少和不易乳化等優(yōu)點(diǎn)[13]。自20世紀(jì)70年代后期商品化產(chǎn)品問(wèn)世以來(lái),SPE技術(shù)發(fā)展迅速,目前已被廣泛應(yīng)用于中藥材中的微量物質(zhì)分析[14-15]。在線SPE以柱切換系統(tǒng)為核心,通過(guò)程序控制多通道切換實(shí)現(xiàn)樣品前處理與分離分析的偶聯(lián),能夠自動(dòng)化實(shí)現(xiàn)從生物樣品前處理到數(shù)據(jù)分析的全部過(guò)程[16-17]。
