HPLC測定槐花藥材不同部位蘆丁和槲皮素含量

  摘要:目的 比較槐花不同部位(槐花、槐米及枝梗)中蘆丁和槲皮素的含量。方法 采用高效液相色譜法進(jìn)行測定,Ultimate XB-C18色譜柱(4.6 mm×250 mm,0.45 ?m),水-甲醇為流動(dòng)相梯度洗脫,流速1 mL/min,柱溫35 ℃,檢測波長256 nm,進(jìn)樣量20 ?L。結(jié)果 蘆丁含量為槐米>槐花>枝梗;槲皮素含量整體較低,且槐花>槐米>枝梗。結(jié)論 槐花各部位蘆丁含量較高,且含量差別不大。槐花中的槲皮素含量明顯高于槐米及其枝梗。

  Abstract:Objective To compare rutin and quercetin content of different parts (flower, bud and pedicel) of Sophora Japonica L.. Methods HPLC method was used with Ultimate XB-C18 column (4.6 mm×250 mm, 0.45 ?m), water and methanol as solvent system with gradient elution. The flow rate was 1 mL/min, column temperature was 35 ℃, detection wavelength was set at 256 nm, and injection volume was 20 ?L. Results The content of rutin was bud>flower>pedicel. The content of quercetin was low on the whole, and in sequence of flower>bud>pedicel. Conclusion In the different parts of Sophora Japonica L., rutin content is high with little difference. Quercetin content of flower is significantly higher than in bud and pedicel.

  槐花為豆科植物槐Sophora japonica L.的干燥花及花蕾,前者稱“槐花”,后者稱“槐米”,具有涼血止血、清肝瀉火的功效[1],F(xiàn)代研究表明,其黃酮類主要成分蘆丁和槲皮素均具有多種藥理作用[2]。近年來,隨著醫(yī)藥工業(yè)的發(fā)展及保健品的開發(fā),槐花作為黃酮類的提取原料,其需求量逐年遞增。但因栽種局限性、生長半野生化及環(huán)保限制采收等原因使槐花產(chǎn)量減少[3]。市售槐花藥材中除了槐花和槐米外,還含有大量的枝梗,在炮制過程中常常被作為非藥用部位而除去[4]。本研究采用高效液相色譜法(HPLC)同時(shí)測定、比較分析同一來源的槐花藥材中的槐花、槐米及其枝梗所含蘆丁及槲皮素的含量,探討槐花同一來源不同部位有效成分的差異性,為槐花藥材的凈制加

  Waters高效液相色譜儀(含1525工作泵及2487檢測器);Adventurer-AR1140型電子分析天平。蘆丁對(duì)照品(中國藥品生物制品檢定所,批號(hào)100080-200707),槲皮素對(duì)照品(中國藥品生物制品檢定所,批號(hào)100081-200907);槐花藥材購于湖南長沙高橋藥材大市場,經(jīng)湖南中醫(yī)藥大學(xué)周日寶教授鑒定為豆科植物槐Sophora japonica L.的干燥花、花蕾及其枝梗;甲醇為色譜純,其余試劑為分析純;液相色譜用水為超純水(自制,0.45 ?m濾膜濾過)。
